Histidine tag (His-tag)
Histidines are very common tags used in the affinity chromatography purification of recombinant proteins. Due to their small size and neutral charge at physiological pH values, his-tags have little interference with the protein structure.
In our design, we added a 6-histidine tag (His-tag) at the N-terminus of our passenger proteins - either cellulase Endo5a or CBD (cellulose binding domain). The passenger proteins were expressed as a cellulolytic complex on the surface of E. coli through AIDA-1 auto-display system.
